Studies on Conditions Affecting the Survival in Vitro of a Malarial Parasite (plasmodium Lophurae)
نویسنده
چکیده
The survival of Plasmodium lophurae in vitro, at temperatures of 39.5-42 degrees C., is favored by a balanced salt solution having a high potassium content, by aeration but not by a very high oxygen tension, by an optimal density of parasites per cubic millimeter, by frequent renewal of the suspending medium, by concentrated red cell extract, by optimal concentrations of plasma or serum, of chick embryo extract, of glucose or glycogen, and of glutathione, and probably by yeast extract and a very low concentration of liver extract. In the best preparations, as judged by infectivity, more than 40 per cent of the parasites were alive on the 3rd day, more than 20 per cent on the 4th day, perhaps 1 per cent on the 5th day, and about 0.05 per cent on the 6th day. Evidence was obtained that the parasites had multiplied during the 1st day of incubation.

On primary structure and biosynthesis of histidine-rich polypeptide from malarial parasite Plasmodium lophurae.
Experiments on the primary structure of a histidine-rich polypeptide isolated from the malarial parasite Plasmodium lophurae indicate that the smaller quantities of amino acids other than histidine form an integral part of the polypeptide and do not arise from a protein contaminating a histidine homopolymer. In culture, the parasites incorporate over 50% of exogenously supplied histidine into t...

The survival of Plasmodium lophurae in vitro is favored by the presence of calcium pantothenate (0.02 mg. per ml). Survival of about 2 weeks in vitro at 40-41 degrees C. has been obtained under the following conditions: a medium consisting of duck red cell extract in balanced salt solution with glutathione and glucose or glycogen, serum, embryo extract, and calcium pantothenate; daily replaceme...
متن کاملStudies on Malarial Parasites Vii. Methods and Techniques Por Cultivation* by Quentin
When Laveran (1) discovered living malarial parasites over 65 years ago in the blood of patients with certain fevers, a new era in the study of malaria was initiated. This discovery of malarial organisms and the subsequent experimental proof by a series of workers that they were the etiological agents of the disease failed, however, to lead to the immediate successful cultivation of the parasit...

The extracellular survival of the malaria parasite Plasmodium lophurae was favored on the 4th day of incubation in vitro by the presence in the medium of added folinic acid at a concentration of about 5 microg. per ml. The development of the human malaria parasite P. falciparum intracellularly in suspensions of human erythrocytes was better in a medium with a high than in one with a low concent...

Plasmodium falciparum, Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are the parasites that infect humans. Plasmodium falciparum and Plasmodium vivax cause most of the malarial infections worldwide. Plasmodium vivax, Plasmodium ovale, Plasmodium malariae, and Plasmodium knowlesi are susceptible to chloroquine. Chloroquine was the world's most widely used antim...
